De Vries, who won the opening round of the 2020 Formula E Season in Saudi Arabia, will share the role with current team-mate Stoffel Vandoorne.
Following his championship winning year in F2 in 2019, de Vries joined the Mercedes-Benz EQ Team in their inaugural season in the all-electric series alongside Vandoorne.
After a dominate lights-to-flag victory to secure his first in the sport, de Vries now leads the Driver's Championship after two rounds while Mercedes currently sit in second in the Team's Championship.
Vandoorne, who previously won the GP2 Championship before driving for McLaren between 2016 and 2018, joined Mercedes as their simulator driver before officially becoming their reserve driver last year.
Taking over from former Haas F1 driver Esteban Gutierrez, de Vries took part in the Abu Dhabi rookie test alongside Vandoorne for Mercedes at the end of last year.
Mercedes, who unveiled their 2021 F1 car today, also commented on the announcement of de Vries to their programme.
A statement said: "We also have a strong reserve driver line-up for the 2021 season, with Stoffel Vandoorne continuing in the role alongside his Mercedes team-mate in Formula E, Nyck de Vries."
